Preparation
This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human S100A6 (rh S100A6; Catalog#10939-H08B; NP_055439.1; Met1-Gly90). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.

Immunofluorescence staining of Human S100A6 in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with Mouse anti-Human S100A6 monoclonal antibody (1:60) at 37℃ 1 hour. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-mouse IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm.
Flow cytometric analysis of Human S100A6 expression on HeLa cells. The cells were treated according to manufacturer’s manual (BD Pharmingen™ Cat. No. 554714), stained with purified anti-Human S100A6, then a FITC-conjugated second step antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.

S100A6 Background Information

S1 proteinis a family of low molecular weight protein found in vertebrates characterized by twoEF-hand calcium-binding motifs. There are at least 21 different S1 proteins, and the name is derived from the fact that the protein is1%soluble in ammonium sulfateat neutralpH. Most S1 proteins are disulfide-linked homodimer, and is normally present in cells derived from theneural crest, chondrocytes, macrophages, dendritic cells, etc. S1 proteins have been implicated in a variety of intracellular and extracellular functions. They are involved in regulation of protein phosphorylation, transcription factors, the dynamics of cytoskeleton constituents, enzyme activities, cell growth and differentiation, and the inflammatory response. S1A6 (S1 calcium binding protein A6) is a member of the S1 family of proteins, and functions in prolactin secretion, and exocytosis. Chromosomal rearrangements and altered expression of S1A6 have been implicated in melanoma.
