Background of Epigenetics

Epigenetics refer to heritable cellular and physiological traits which are not linked to changes of DNA sequences. Although each cell contains the same DNA sequences, epigenetic regulation control the correct transcriptional pattern in a given cell type. Therefore, epigenetic regulation plays an important role in cell proliferation, differentiation and development.
Epigenetic modifications include histone modification, DNA methylation and RNAi. For post-translational modifications of histone proteins, there are histone acetylation, methylation, phosphorylation, ubiquitylation and sumoylation. Histone acetylation and methylation are the two most abundant and widely studied histone modifications, and are highly regulated by multiple proteins. 'Writers' add methyl or acetyl group onto histones, 'erasers' remove these marks and 'readers' recognize modified histones. For example, histone acetylation is regulated by two families of enzymes, histone acetyltransferases (HATs) to add acetyl group onto lysine residues on histones and histone deacetylases (HDACs) for removal. Histones with modified lysine residues are recognized by bromodomain-containing family of proteins. Since epigenetics are implicated in fundamental processes, aberrant epigenetic regulation causes altered gene expression which may lead to disease state. Epigenetics is a promising area for drug discovery.
